比较研究补阳还五汤及其有效部位组方对脑缺血再灌注后脑组织兴奋性氨基酸 (EAA)含量和海马CA1区超微结构的影响。采用沙鼠脑缺血模型 ,缺血 15min后进行再灌注 ,于再灌注不同时间取脑组织测定。结果 :①在再灌注 48h ,补阳还五汤及其有效部位方可使脑组织谷氨酸 (Glu)含量降低 ,有效部位方还可使天冬氨酸 (Asp)含量降低 ;②在再灌注后 5d ,补阳还五汤及其有效部位方均可对抗海马CA1区神经元变性死亡 ,对CA1区超微病理改变有改善作用 ,两者改善程度相近 ;③在再灌注后 5d ,补阳还五汤及其有效部位方均可减少海马CA1区神经元凋亡发生的例数 ,具有抗脑缺血后神经元凋亡的作用 ,两者作用相近。认为补阳还五汤及其有效部位方均具有抗缺血性脑损伤的作用 ,它可对抗EAA的兴奋毒性 ,改善神经元的超微病理结构 ,对抗神经元凋亡。
To compare the effects of Buyang Huanwu Decoction and its effective fraction on the contents of excitatory amino acid (EAA) in brain tissue and the ultrastructure of hippocampal CA1 area after cerebral ischemia-reperfusion in rats. Using gerbil cerebral ischemia model, reperfusion was performed 15 min after ischemia, and brain tissue was taken at different time after reperfusion. RESULTS: 1 After 48 hours of reperfusion, Buyang Huanwu Decoction and its effective fractions can reduce the content of glutamate (Glu) in brain tissue, and the content of aspartic acid (Asp) in the effective site can also be reduced. 2 After 5 days of instillation, Buyang Huanwu Decoction and its effective positions can all reverse the degeneration and death of neurons in the hippocampal CA1 region, and have an improvement effect on the ultrastructural pathological changes in the CA1 region. The improvement degree of the two is similar; 3 at 5 days after reperfusion, Yang Huanwu Decoction and its effective positions can reduce the number of neurons apoptosis in hippocampal CA1 region and have the effect of anti-apoptosis after cerebral ischemia. The two effects are similar. It is considered that Buyang Huanwu Decoction and its effective sites have anti-ischemic brain damage, which can counteract the excitotoxicity of EAA, improve the ultrastructural pathology of neurons, and counter neuronal apoptosis.
