目的 :探讨端粒酶抑制剂对动物在体肿瘤的治疗作用及对放射治疗的增敏价值。方法 :将 SGC- 790 1胃癌细胞接种 BAL B/ c小鼠 ,采用端粒酶抑制剂 AZT联合放疗治疗小鼠胃癌 ,观察其对肿瘤体积和肿瘤端粒酶活性的影响。结果 :未治疗组小鼠肿瘤体积增加了 2 .5倍 ,而 AZT组、放疗组及 AZT联合放疗组肿瘤体积分别缩小了 2 6 .3%、36 .8%及 70 .5 %。肿瘤端粒酶活性检测 ,未治疗组、AZT组、放疗组及 AZT联合放疗组的相对端粒酶活性分别为 10 0、5 5 .2、2 4 .5及 12 .1。结论 :AZT、放疗均能使肿瘤体积缩小并降低端粒酶活性 ,且 AZT能增加肿瘤的放射敏感性。
Objective: To investigate the therapeutic effect of telomerase inhibitor on tumor in vivo and its sensitization value to radiation therapy. Methods: BALB / c mice were inoculated with gastric cancer cell line SGC-790 1 and the gastric cancer was treated with telomerase inhibitor AZT combined with radiotherapy. The effects of telomerase inhibitor AZT and telomerase on the tumor volume and tumor telomerase activity were observed. Results: The tumor volume of untreated mice increased by 2.5 times, while the tumor volume in AZT group, radiotherapy group and AZT combined with radiotherapy group decreased by 26.3%, 36.8% and 70.5% respectively. The relative telomerase activities of tumor telomerase activity test, untreated group, AZT group, radiotherapy group and AZT combined radiotherapy group were 10 0,5 5,2,2 4 5 and 12 1, respectively. Conclusion: Both AZT and radiotherapy can reduce tumor volume and decrease telomerase activity, and AZT can increase tumor radiosensitivity.
