目的:通过观察调中汤对脾虚肝旺型功能性消化不良幼鼠胃肠动力的影响,探讨调中汤对脾虚肝旺型功能性消化不良的具体作用机制。方法:将清洁级SD幼鼠36只,日龄21~28 d,体质量50~70 g,随机分为6组,每组6只:空白对照组、模型组、调中汤高剂量组、调中汤中剂量组、调中汤低剂量组、多潘立酮组(西药组)。幼鼠在购入后适应环境喂养1周,正常喂养,第2周开始,除对照组每日常规饲养不予造模外,其余组采用不规则喂养、饮用4℃冰水及夹尾刺激法造模。造模成功后,从造模后第7天开始用药,持续给药14 d。各组幼鼠在末次给药前12 h禁食(自由饮水),1 min内处死动物,同时取出胃及小肠。计算胃排空率及小肠推进速度。结果:调中汤高、中、低剂量组幼鼠胃排空率明显升高,较模型组比较(P<0.01);调中汤高剂量组能明显促进幼鼠小肠推进比,与模型组比较(P<0.01)。结论:成功建立脾虚肝旺型功能性消化不良幼鼠模型,并观察到调中汤能明显提高脾虚肝旺型功能性消化不良幼鼠胃排空率、小肠推进速度。与西药组比较,调中汤高剂量组效果更好。说明提高幼鼠胃排空率、小肠推进比是调中汤治疗脾虚肝旺型FD的作用机制之一。
Objective: To observe the effect of Tiaozhong Decoction on gastrointestinal motility in young rats with functional dyspepsia due to spleen-deficiency syndrome and to explore the specific mechanism of tiaozhong decoction on functional dyspepsia with spleen-deficiency syndrome. Methods: Thirty-six clean SD rats, aged from 21 to 28 days and body weight ranging from 50 to 70 g, were randomly divided into 6 groups with 6 rats in each group: blank control group, model group, TZT high dose group, Tonic medium dose group, Tonic medium low dose group, domperidone group (western medicine group). Young rats were fed to the environment for 1 week after feeding, normal feeding, the beginning of the second week, except for the control group daily routine feeding without modeling, the other group using irregular feeding, drinking 4 ℃ ice water and tail tails stimulation Modeling. After the success of modeling, starting from the 7th day after model administration, continuous administration for 14 days. The pups of each group were fasted (freely drinking water) 12 h before the last administration, animals were sacrificed within 1 min, and the stomach and small intestine were removed simultaneously. Calculate the rate of gastric emptying and intestinal propulsion speed. Results: Compared with the model group, Tiaozhong decoction high dose group could significantly promote the small intestine to promote the ratio of rats to the model group (P <0.01). Conclusion: The model of functional dyspepsia in spleen-deficiency liver-wu-type mice was successfully established. It was observed that Tiaoxin decoction could significantly improve gastric emptying rate and small intestine propulsion rate in spleen-deficiency liver dysfunction-induced dyspepsia rats. Compared with Western medicine group, Tiaozhong Tang high dose group is better. It is indicated that improving the rate of gastric emptying in rats and promoting the small intestine is one of the mechanisms of Tiaozhong Decoction in treating FD with spleen deficiency.
