目的 :建立高效液相色谱法测定通降胶囊中大黄的大黄素、大黄酚和大黄酸含量。方法 :采用VARLAG公司C18柱(4.6mm× 2 5 0mm ,5 μm) ;流动相 :甲醇 - 0 .1%磷酸溶液 (85∶15 ) ,流速 1.2ml·min-1,检测波长 :4 4 0nm。结果 :大黄素在32~ 16 0ng、大黄酚在 80~ 4 0 0ng、大黄酸在 112~ 5 6 0ng范围内 ,进样量 (ng)与峰面积呈良好的线性关系。平均回收率分别为 99.2 % (大黄素 ,RSD =2 .96 % ) ,97.4 % (大黄酚 ,RSD =1.5 9% ) ,10 2 .9% (大黄酸 ,RSD =2 .4 0 % )。结论 :本法快速、简便、准确。
Objective : To establish a HPLC method for the determination of emodin, chrysophanol and rhein in Rhododendron capsules. METHODS: VARLAG C18 column (4.6 mm × 250 mm, 5 μm) was used; mobile phase: methanol - 0.1% phosphoric acid solution (85:15), flow rate 1.2 ml · min-1, detection wavelength: 4 4 nm . Results: The emodin ranged from 32 to 160 ng, chrysophanol from 80 to 400 ng, and rhein from 112 to 560 ng. The injection volume (ng) showed a good linear relationship with the peak area. The average recoveries were 99.2% (emodin, RSD = 2.96%), 97.4 % (chryphenol, RSD = 1.59%), 102.9% (rhein, RSD = 2.4%). Conclusion : This method is fast, simple and accurate.
