目的:探讨原发性高血压(EH)患者颈总动脉超声等临床变量与髓过氧化物酶(MPO)基因变异的关系。方法:应用聚和酶链反应(PCR)检测EH组107例及对照组97例患者MPO基因型,比较不同基因型之间的分布频率。同时使用B型彩色多普勒超声等检测2组患者的颈总动脉超声及其他临床变量,并进行对照分析。结果:正常人群GG、GA、AA基因型频率分别为56.7%、40.2%和3.1%,EH病例组分别为70.1%、29.0%和0.9%,EH组MPO基因AA+GA基因型频率低于对照组,二者比较有差异性(P=0.047)。EH患者的颈总动脉内径、内膜厚度等临床变量的变化与MPO不同基因型相关。结论:本研究人群MPO基因多态性与EH遗传易感性相关。EH患者的颈总动脉内径、内膜厚度及各临床变量的变化与MPO基因463G/A的变异有关。
Objective: To investigate the relationship between the clinical variables such as common carotid artery and the myeloperoxidase (MPO) gene variation in patients with essential hypertension (EH). Methods: MPO genotypes of 107 patients in EH group and 97 patients in control group were detected by polymerase chain reaction (PCR), and the distribution frequencies of different genotypes were compared. At the same time using B-mode color Doppler ultrasound detection of two groups of patients with common carotid artery ultrasound and other clinical variables, and control analysis. Results: The frequencies of genotypes of GG, GA and AA were 56.7%, 40.2% and 3.1% respectively in normal subjects, 70.1%, 29.0% and 0.9% in EH cases, respectively. The frequencies of AA + GA genotypes in MPO gene of EH group were lower than those in control subjects Group, the two were quite different (P = 0.047). Changes in clinical variables such as carotid artery internal diameter and intima thickness in patients with EH correlated with different MPO genotypes. CONCLUSIONS: The MPO gene polymorphisms in this study population were associated with genetic susceptibility to EH. The changes of carotid artery diameter, intima thickness and clinical variables in EH patients were related to the variation of MPO gene 463G / A.
