目的:观察强肌健力口服液与龟板混合血清对大鼠骨髓间充质干细胞(MSCs)体外增殖的影响。方法:使用密度梯度离心法分离大鼠MSCs,5-溴脱氧嘧啶(Brdu)和CD44双重免疫组化鉴定,传3代后的MSCs分为强肌健力口服液与龟板混合血清组和空白血清组、强肌健力口服液组,四甲基偶氮唑盐(MTT)法检测各组MSCs的生长情况。结果:强肌健力口服液与龟板混合血清各组均能促进MSCs的增殖,与空白血清组比较P均<0.01,强肌健力口服液与龟板中剂量混合血清组5μl+95μl/ml、25μl+75μl/ml、50μl+50μl/ml与强肌健力口服液组比较,P<0.01。结论:脾肾并补较之单纯补脾更能促进MSCs的生长。
OBJECTIVE: To observe the effect of Qiangji Jianli oral liquid and turtle-plate mixed serum on proliferation of rat bone marrow mesenchymal stem cells (MSCs) in vitro. METHODS: The rat MSCs were isolated by density gradient centrifugation and identified by double immunohistochemistry using Brdu and CD44. Three generations of MSCs were divided into the group consisting of Qiangji Jianli oral liquid and turtle plate mixed serum group and blank serum. The growth of MSCs in each group was detected by MTT assay and MTT group. Results: Qiangji Jianli oral liquid combined with turtle shell mixed serum could promote the proliferation of MSCs, P<0.01 compared with blank serum group, Qiangji Jianli oral liquid and turtle shell medium dose mixed serum group 5μl+95μl/ml, 25μl+75μl/ml, 50μl+50μl/ml compared with Qiangji Jianli oral solution group, P<0.01. Conclusion: The spleen and kidney supplementation can promote the growth of MSCs more than simply supplementing the spleen.
