清肺口服液对RSV感染哮喘急性发作小鼠肺组织炎症及气道反应性的影响

目的:探讨清肺口服液对呼吸道合胞病毒(RSV)感染哮喘急性发作小鼠模型肺组织炎症及气道反应性的影响。方法:60只雄性6~8周龄Balb/C小鼠,每组10只,随机分为正常组,模型组,清肺口服液高、中、低剂量组(2.34,1.17,0.59 g·kg-1·d-1),利巴韦林组(0.1 g·kg-1·d-1),共6组,卵细胞(OVA)致敏及雾化激发哮喘模型,经RSV隔日滴鼻连续3次感染,获得RSV感染哮喘模型;治疗组予清肺口服液分高、中、低剂量ig;正常予等量生理盐水ig:阳性药组给予利巴韦林注射液ip。末次激发24 h后,用Buxco RC系统检测小鼠气道反应性;收集支气管肺泡灌洗液(BALF)进行嗜酸细胞分析;肺组织经HE染色,PAS染色,VG染色评估炎症反应。结果:与正常组比较,模型组小鼠BALF中嗜酸粒细胞比例(EOS%)明显升高,气道反应性较正常组明显增高(P<0.01),模型组小鼠肺组织呈现哮喘合并感染典型改变,组织充血水肿,分泌物显著增多;气道周围炎性细胞浸润、胶原增生显著;与模型组比较,清肺口服液中剂量组BALF中EOS%明显降低,清肺口服液及利巴韦林干预均可显著改善气道高反应性(P<0.05),其中中剂量清肺口服液效果尤为显著,气道功能与正常组相比差异无显著性,清肺口服液中剂量可以明显减轻小鼠肺组织充血水肿及炎性细胞浸润,气道分泌物及气道周围胶原增生较模型组明显改善。结论:中剂量清肺口服液可以明显减轻RSV感染哮喘急性发作小鼠肺组织炎症及气道高反应性。 Objective: To investigate the effect of Qingfei Oral Liquid on lung inflammation and airway responsiveness in asthmatic mice with respiratory syncytial virus (RSV) infection. Results: Compared with the normal group, the eosinophil ratio (EOS%) of BALF in model group was significantly increased and the airway reactivity was significantly higher than that in normal group (P <0.01). The lung tissue of model group showed asthma combined The typical changes of infection, tissue congestion and edema, secretions increased significantly; inflammatory cell infiltration around the airways, collagen hyperplasia significantly; compared with the model group, Qingfei oral liquid dose group BALF EOS% was significantly reduced, Qingfei oral liquid and Lee The intervention of ribavirin can significantly improve the airway hyperresponsiveness (P <0.05), of which the effect of medium dose Qingfei oral liquid is particularly significant, airway function was no significant difference compared with the normal group, Qingfei oral liquid dose Significantly reduce mouse lung congestion and edema and inflammatory cell infiltration, airway secretions and airway proliferation of collagen than the model group was significantly improved. Conclusion: The medium-dose Qingfei oral liquid can significantly reduce lung inflammation and airway hyperresponsiveness in RSV-infected acute asthma mice.
