Effect of host Prdx6 on Intracellular Survival of Brucella

  Brucellosis is a worldwide zoonotic infectious disease and transmitted through various ways,which leads to a great harm to human and animals.The pathogen of brucellosis is Brucella,which mainly residents in macrophage cells,survives and replicates in host cells.However,the mechanisms of Brucella surviving in macrophage cells have not been fully understood.Peroxiredoxin 6(Prdx6)is a bifunctional protein,which shows not only the GSH peroxidase activity but the phospholipase A2 activity,and plays an important role against oxidative damages and in regulating apoptosis.Here,the recombinant mouse(Mus musculus)Prdx6(MmPrdx6)was expressed and purified,and the monoclonal antibodies against MmPrdx6 were prepared.Using Brucella suis S2 strain to challenge the murine macrophage cell line RAW264.7,the level of intracellular Prdx6 expression first decreased and then increased.Overexpressing Prdx6 in macrophages resulted in the increase of Brucella in RAW264.7 cells,and knocking it down reduced Brucella in cells.Therefore,host Prdx6 can enhance the intracellular survival of Brucella and plays an important role in Brucella infection.
