构成核小体的组蛋白上携带大量的表观遗传信息,其中组蛋白的翻译后修饰是最重要的形式之一.这些修饰影响以DNA为模板的几乎所有过程,如基因表达、DNA复制等.四膜虫的EZL2基因编码一种组蛋白赖氨酸N-甲基转移酶,可催化形成H3组蛋白第27位赖氨酸的二甲基化和三甲基化(H3K27me2/H3K27me3)[1],但EZL2蛋白对四膜虫基因表达的具体作用和调控机制仍不清晰.本工作对四膜虫野生型和△EZL2株系进行了表型分析,同时采用微球藻核酸内切酶(Micrococcal nuclease,MNase)处理及蔗糖密度梯度离心提取高纯度单核小体,并进行了Illumina测序.
