[Objective]Optimizing an efficient system is the basis of related research by using in situ hybridization.[Method]In this study,based on 3 reported probe sequence of tdh,trh and toxR,methods of in situ hybridization of pathogenic Vibrio parahaemolyticus have been optimized,including pretreatment of colonies,se-lection of fixation method,temperature and duration of hybridization and duration of blocking.[Results]The high specific,low background and distinctly colored results have been acquired by the optimized methods used 2h for crosslinking at 80 ℃,8h for hybridization at 38℃ and 2h for blocking.[Conclusion]This study creates an important platform to following colony in situ hybridization research,provides the technical support for the detection of pathogenic and nonpathogenic strains.
