Estrogen but not ERβ, is positively expressed in cancer associated (ER Receptor in CAF fibroblast (CAF) of prostate cancer (PCa).However, the impact of ER cells to control PCa cell growth remains unclear.We found here that addition of ERα in CAF cells led to enhance PC3 cells growth in MTT assay.Similar results were also obtained when we replaced PC3 cells with other PCa cells including LNCaP, 22RV1 or C4-2 cells.The results from 3D colony formation assay also showed that addition of ERα in CAF led to enhance colony formation in CWR22RV1 and PC3 cells.Using in vivo + mouse model with orthotopic co-implantation of CWR22RV1 cells with CAF-ER vs-cells, we also with CAF-ER confirmed the in vitro co-culture data showing the mice in the CAF ER+ co-implanted PCa group had stronger IVIS signals with bigger tumor size than the mice in the CAF ER-group.Together, our results from in vitro co-culture and in vivo mouse model all proved that CAF ER might play positive roles to enhance the PCa growth.
