以取自巴西橡胶树(Hevea brasiliensis Muell.Arg.)种子实生苗的腋芽为外植体,利用含1.0mg/LKT,1.0mg/L2,4-D,20g/L蔗糖,以及4g/L琼脂粉的MS培养基成功获得腋芽无菌苗。将其转入附加5.0mg/LNAA,3.0mg/LIBA,50g/L蔗糖,以及4g/L琼脂粉的MS培养基可进一步诱导生根。试验中我们还发现,尽管从叶片及腋芽外植体均能诱导出大量愈伤组织,但最终均未能实现体细胞胚胎发生及植株再生。采用合适的培养基能够诱导腋芽外植体离体萌发并生根,这一结果表明,通过进一步的研究和探索,完全有可能借助该途径建立巴西橡胶树的高效离体微繁体系。
Axillary buds from seedlings of Hevea brasiliensis Muell. Arg. Seedlings were used as explants with 1.0 mg / L KT, 1.0 mg / L 2,4-D, 20 g / L sucrose and 4 g / MS medium successfully obtained axillary bud sterile seedlings. Rooting was further induced by transfer to MS medium supplemented with 5.0 mg / L NAA, 3.0 mg / L IBA, 50 g / L sucrose, and 4 g / L agarose. In the experiment, we also found that although a large amount of callus can be induced from leaf explants and axillary bud explants, ultimately all failed to achieve somatic embryogenesis and plant regeneration. Appropriate culture medium can induce axillary bud explants in vitro germination and rooting, the results show that, through further research and exploration, it is entirely possible to use this approach to establish high efficient micropropagation system of rubber tree in Brazil.
