强骨胶囊治疗维甲酸所致骨质疏松大鼠的疗效观察

目的:评价强骨胶囊对维甲酸所致骨质疏松大鼠的药效。方法:取3月龄雌性大鼠40只,随机分为造模组24只与空白组16只,造模组按80 mg/kg灌服1%CMC-Na配成的维甲酸混悬液,同时空白组灌服等体积生理盐水,连续灌药2周后,于两组随机取8只大鼠进行模型评价。造模成功后,将造模组大鼠随机分为生理盐水组和强骨胶囊组各8只。强骨胶囊组按67.5 mg/kg强骨胶囊灌胃,生理盐水组按等体积生理盐水灌胃,连续治疗4周后取材检测。结果:造模2周后,与空白组相比,模型组骨体积分数(P<0.05)、骨小梁数目(P<0.01)均明显减少,骨小梁分离度明显增大(P<0.01),说明维甲酸造模2周后大鼠出现骨质疏松;给药4周后,相比于生理盐水组,给药后强骨胶囊组大鼠体重逐渐增高,在给药2周、4周时较明显(P<0.05);Mirco-CT检测显示,与生理盐水组比较,强骨胶囊组第5腰椎骨体积分数(P<0.01)、骨小梁数目(P<0.05)以及骨小梁厚度(P<0.05)均明显升高,骨小梁分离度显著降低(P<0.05);生物力学检测结果显示,与生理盐水组比较,强骨胶囊组股骨最大载荷与弹性模量均明显增高(P<0.01);血清学检测结果显示,与生理盐水组比较,强骨胶囊组血清25-HVD表达水平明显升高(P<0.05),Ca(P<0.05)、Mg(P<0.01)、β-CTx(P<0.05)和ALP(P<0.05)表达水平均明显降低。结论:强骨胶囊能有效治疗维甲酸所致的大鼠骨质疏松症。 Objective: To evaluate the efficacy of Qianggu capsule on retinoic acid-induced osteoporosis in rats. Methods: Forty female 3-month-old rats were randomly divided into 24 rats in model group and 16 rats in blank group. The rats in model group were dosed with 1% CMC-Na retinoic acid suspension at 80 mg / kg, At the same time, the blank group was given an equal volume of normal saline. After two weeks of continuous infusion, eight rats were randomly selected from the two groups for model evaluation. After successful modeling, rats in model group were randomly divided into normal saline group and Qiangu capsule group, each with 8 rats. Qiangu capsule group was treated with Qiangu capsule (67.5 mg / kg). The rats in normal saline group were given gavage with equal volume of normal saline. After continuous treatment for 4 weeks, samples were taken for detection. Results: Compared with the blank group, the bone volume fraction (P <0.05) and trabecular number (P <0.01) in the model group decreased significantly and the trabecular separation increased significantly (P <0.01) ), Indicating that retinoic acid in rats after 2 weeks of osteoporosis; 4 weeks after administration, compared with the saline group, Qiangu capsule after administration of rats gradually increased body weight, after 2 weeks of administration, 4 Compared with the saline group, the bone volume fraction of the 5th lumbar vertebrae of Qiangu capsule group (P <0.01), the number of trabecular meshwork (P <0.05) and the size of small bone The beam thickness (P <0.05) were significantly increased, and the trabecular separation was significantly reduced (P <0.05). Biomechanical test results showed that the maximum load and elastic modulus of Qiangu capsule group were significantly higher than that of saline group (P <0.01). The results of serological tests showed that the expression of 25-HVD in Qiangu capsule group was significantly higher than that in the normal saline group (P <0.05) ), Β-CTx (P <0.05) and ALP (P <0.05). Conclusion: Qianggu capsule can effectively treat rat osteoporosis caused by retinoic acid.
