目的 研究外源性p1 6基因对肺癌细胞生物学特性的影响。方法 利用脂质体介导的基因转染方法将外源性p1 6基因转入p1 6基因缺陷的人肺腺癌细胞株A54 9中 ,用逆转录聚合酶链反应 (RT PCR)及免疫组化的方法检测p1 6基因的表达 ,同时观察转染后细胞恶性生长的变化。结果 外源性p1 6基因在A54 9细胞中能稳定表达 ,转染后A54 9细胞生长速度明显减慢 ,在软琼脂上形成克隆的能力降低。流式细胞仪检测显示A54 9细胞G1期阻滞并发生了凋亡 ,接种裸鼠后致瘤性降低 ,肿瘤生长明显减慢。结论 外源性p1 6基因导入人肺癌细胞株A54 9中可稳定表达并抑制细胞的恶性生长 ,同时诱导凋亡。
Objective To study the effect of exogenous p16 gene on the biological characteristics of lung cancer cells. Methods The liposome-mediated gene transfection method was used to transfer the exogenous p16 gene into p16-deficient human lung adenocarcinoma cell line A549, using reverse transcription polymerase chain reaction (RT-PCR) and immunization. The method was used to detect the expression of p16 gene and observe the malignant growth of cells after transfection. Results The exogenous p16 gene was stably expressed in A54 9 cells. After transfection, the growth rate of A549 cells was significantly slower, and the ability to form clones on soft agar was reduced. Flow cytometry showed that A54 9 cells arrested in G1 phase and developed apoptosis. After inoculation of nude mice, tumorigenicity was decreased and tumor growth was significantly slowed down. Conclusion The introduction of exogenous p16 gene into human lung cancer cell line A54 9 can stably express and inhibit the malignant growth of cells and induce apoptosis.
