作者用自行研制的鼠源性胃癌细胞单克隆抗体与溴化氰活化的葡聚糖4B(CNBr-Sepharose4B)偶联,从胃癌组织匀浆中分离相关抗原(gastriccarcinomarelatedantigen,GCRA)。通过免疫家兔获得兔源性多抗,建立酶免疫GCRA测定法。用以检测167例胃液标本,结果胃癌组阳性率71.9%(41/57);癌前期病变28.6%(6/21);正常组6.6%(6/89)。胃癌组与正常组比较P<0.001。GCRA对胃癌诊断敏感性89.1%,特异性71.9%,提示检测胃液GCRA可提高胃癌的检出率。
The authors used a self-developed monoclonal antibody against mouse gastric cancer cells coupled with cyanogen bromide-activated dextran 4B (CNBr-Sepharose 4B) to isolate relevant antigens (GCRA) from gastric cancer tissue homogenates. A rabbit-derived polyclonal antibody was obtained by immunizing rabbits and an enzyme immunometric GCRA assay was established. For the detection of 167 gastric fluid specimens, the positive rate of gastric cancer group was 71.9% (41/57); the precancerous lesion was 28.6% (6/21); the normal group was 6.6% (6/89). Compared with normal group, gastric cancer group P<0.001. GCRA has a sensitivity of 89.1% and a specificity of 71.9% in the diagnosis of gastric cancer, suggesting that GCRA can improve the detection rate of gastric cancer.
