目的探讨白细胞介素37(IL-37)对小鼠脾脏树突状细胞(DC)运动和迁移能力的影响。方法应用免疫磁珠细胞分选法分离小鼠脾脏内的DC,流式细胞术鉴定细胞纯度,激光共聚焦显微镜连续拍摄细胞,Volocity软件分析DC运动速度,基因芯片分析C趋化因子受体1(XCR1)的mRNA表达水平,聚合酶链式反应和流式细胞术验证芯片结果,TranswellTM方法检测DC的迁移能力。结果免疫磁珠能分离获得纯度>95%的DC,IL-37降低细胞的运动速度,减少XCR1的表达,抑制DC的迁移。结论 IL-37抑制DC的运动和迁移。
Objective To investigate the effect of interleukin-37 (IL-37) on the motility and migration of dendritic cells (DCs) in mice. Methods DCs in spleen of mice were isolated by magnetic activated cell sorting (FCM). Cell purity was determined by flow cytometry. Cells were continuously photographed by laser confocal microscopy. The velocity of DCs was analyzed by Volocity software. The expression of C chemokine receptor 1 (XCR1) mRNA expression, polymerase chain reaction and flow cytometry to verify the chip results, TranswellTM method to detect DC migration. Results Immunomagnetic beads were able to separate DCs with purity> 95%. IL-37 could reduce the speed of cell movement, reduce the expression of XCR1 and inhibit the migration of DCs. Conclusion IL-37 inhibits DC motility and migration.
