目的为研发天然抗癌活性产物新资源,丰富药用植物内生真菌代谢产物多样性。方法从马比木植物中分离获得一株木霉属内生真菌Trichoderma sp.编号为GX-8,对其发酵产物进行分离纯化后得到一种甾体类化合物,以人非小细胞肺癌细胞株A549和人前列腺癌细胞株PC3为体外抗肿瘤筛选模型,采用MTT法对目标化合物进行抗癌活性筛选。结果该化合物体外对两种癌细胞的增殖具有一定抑制作用,IC_(50)值分别为37.35μmol·L~(-1)和50.83μmol·L~(-1)。结论其效果优于阳性对照组盐酸阿霉素,且呈现剂量-药效依赖关系,可作为一种微生物来源的抗癌活性产物新资源。
The purpose is to develop new resources of natural anti-cancer active products and enrich the diversity of metabolites of endophytic fungi in medicinal plants. Methods A strain of Trichoderma sp. Trichoderma sp. No. GX-8 was isolated from the plants of Mabi and was isolated and purified from the fermentation product to obtain a steroidal compound. The human non-small cell lung cancer cell line A549 and human prostate cancer cell line PC3 as in vitro anti-tumor screening model, the target compounds were screened for anti-cancer activity by MTT method. Results The compound could inhibit the proliferation of both cancer cells in vitro. The IC 50 values were 37.35μmol·L -1 and 50.83μmol·L -1, respectively. Conclusion The result is better than that of the positive control group, and it shows a dose-effect dependent relationship, which can be used as a new resource of anticancer activity product derived from microorganism.
