目的探讨心脏肥大细胞在自发性高血压大鼠(SHR)心肌重构中的作用。方法应用病理检查、计算机分析结合逆转录聚合酶链式反应等方法,观察SHR及Wistar-Kyoto大鼠(WKY)收缩压、左室重量指数、心肌细胞直径、肥大细胞密度、心肌胶原容积分数(CVF)、心肌血管周围胶原面积比(PV-CA)和心肌Ⅰ、Ⅲ型胶原mRNA表达水平的变化。肥大细胞密度与左室重量指数、CVF及PVCA之间的关系采用相关分析。结果与WKY比较,SHR收缩压为(206±18)比(108±10)mm Hg(P<0.01);SHR组左室重量指数为(4.6±0.4)比(3.3±0.3)mg/g,(P<0.01);SHR组心肌细胞长径为(17.4±1.9)比(10.0±2.2)μm(P<0.01);SHR组心肌细胞短径为(9.0±2.0)比(5.8±1.7)μm(P<0.01);SHR组肥大细胞密度为(7.4±3.2)比(1.9±1.2)个/mm2(P<0.01),SHR组肥大细胞密度为WKY组的3.9倍。SHR组CVF、PVCA分别为46.4%±7.8%和1.9±0.9,WKY组分别为24.4%±10.7%和0.4±0.1,SHR组明显升高(P<0.01)。SHR组心肌Ⅰ、Ⅲ型胶原mRNA表达相对含量也均明显高于WKY(P<0.01),心脏肥大细胞密度与左室重量指数、CVF及PVCA存在明显的正相关(相关系数分别为0.67、0.87和0.95,P<0.01)。结论心脏肥大细胞密度增加可能是促进SHR心肌重构的重要原因。
Objective To investigate the role of cardiac mast cells in myocardial remodeling in spontaneously hypertensive rats (SHR). Methods The systolic blood pressure, left ventricular mass index, myocardial cell diameter, mast cell density and myocardial collagen volume fraction (WKY) were observed by pathological examination, computer analysis and reverse transcription polymerase chain reaction CVF), myocardial perivascular collagen area ratio (PV-CA) and myocardial collagen type Ⅰ, Ⅲ mRNA expression changes. Mantle cell density and left ventricular mass index, CVF and PVCA relationship between the use of correlation analysis. Results Compared with WKY, SHR had a systolic blood pressure of (206 ± 18) vs (108 ± 10) mm Hg (P <0.01) and a mean of 4.6 ± 0.4 (3.3 ± 0.3) mg / (P <0.01). The length of myocardial cells in SHR group was (17.4 ± 1.9) vs (10.0 ± 2.2) μm (P <0.01) (P <0.01). The density of mast cells in SHR group was (7.4 ± 3.2) vs (1.9 ± 1.2) / mm2 (P <0.01), and that of SHR group was 3.9 times that of WKY group. The levels of CVF and PVCA were 46.4% ± 7.8% and 1.9 ± 0.9 in SHR group, 24.4% ± 10.7% and 0.4 ± 0.1 in WKY group, respectively, and significantly higher in SHR group (P <0.01). The relative contents of myocardial collagen type Ⅰ and type Ⅲ mRNA in SHR group were also significantly higher than those in WKY group (P <0.01). There was a positive correlation between cardiac mast cell density and LV mass index, CVF and PVCA (correlation coefficients were 0.67,0.87 And 0.95, P <0.01). Conclusion The increased density of cardiac mast cells may be an important reason to promote myocardial remodeling in SHR.
