目的通过分析LGR5、SALL4、NKD1在乳腺癌组织与癌旁乳腺组织中的表达水平,探讨三者在乳腺癌组织中的表达及其相关性。方法选择2015年6月至2016年9月内蒙古包钢医院确诊并行乳腺癌切除术患者的乳腺癌组织30例及其对应的癌旁乳腺组织30例,年龄36~78岁,平均年龄57岁。应用Western blot从蛋白水平检测SALL4、LGR5、NKD1在乳腺癌组织及癌旁乳腺组织中的表达。结果 LGR5、SALL4、NKD1蛋白在乳腺癌组织表达分别为0.63±0.14、0.67±0.13、0.51±0.16(P<0.05),癌旁乳腺组织表达分别为0.53±0.16、0.58±0.11、0.61±0.11(P<0.05)。乳腺癌组织中LGR5与SALL4蛋白呈正相关(r=0.402,P<0.05);LGR5与NKD1蛋白呈负相关(r=-0.685,P<0.05);SALL4与NKD1蛋白呈负相关(r=-0.370,P<0.05)。LGR5、SALL4、NKD1蛋白在TNM分期中Ⅰ+Ⅱ期组表达分别为0.59±0.13、0.64±0.13、0.55±0.17(P<0.05),在Ⅲ期组中表达分别为0.75±0.10、0.75±0.09、0.41±0.11(P<0.05)。LGR5、SALL4、NKD1蛋白在淋巴结转移组中表达分别为0.73±0.11、0.74±0.08、0.40±0.11(P<0.05),在淋巴结未转移组中表达分别为0.58±0.13、0.64±0.14、0.57±0.16(P<0.05)。LGR5、SALL4、NKD1蛋白与乳腺癌患者的年龄、肿瘤的大小、肿瘤部位无关(P>0.05)。结论 LGR5、SALL4和NKD1的异常表达与乳腺癌发生、发展、浸润及转移密切相关,三者的联合检测可能对评估乳腺癌的生物学行为具有重要意义。LGR5、SALL4和NKD1可能是导致乳腺癌的发生、发展的重要因子。
OBJECTIVE: To investigate the expression of LGR5, SALL4 and NKD1 in breast cancer tissues and adjacent normal breast tissues, and to explore their correlation with breast cancer. Methods Thirty cases of breast cancer and 30 corresponding paracancer breast tissues from June 2015 to September 2016 in Baotou Steel Hospital of Inner Mongolia were enrolled in this study. The average age was 57 years (range, 36-78 years). Western blot was used to detect the expression of SALL4, LGR5 and NKD1 in breast cancer tissues and adjacent normal breast tissues. Results The expressions of LGR5, SALL4 and NKD1 in breast cancer tissues were 0.63 ± 0.14, 0.67 ± 0.13 and 0.51 ± 0.16 respectively (P <0.05), while the expressions of LGR5, SLE4 and NKD1 in breast cancer tissues were 0.53 ± 0.16, 0.58 ± 0.11 and 0.61 ± 0.11 P <0.05). There was a positive correlation between LGR5 and SALL4 protein in breast cancer (r = 0.402, P <0.05); LGR5 was negatively correlated with NKD1 protein (r = -0.685, P <0.05); SALL4 was negatively correlated with NKD1 protein , P <0.05). The expressions of LGR5, SALL4 and NKD1 protein in stage Ⅰ + Ⅱ of TNM stage were 0.59 ± 0.13,0.64 ± 0.13 and 0.55 ± 0.17 respectively (P <0.05), and in stage Ⅲ group they were 0.75 ± 0.10,0.75 ± 0.09 , 0.41 ± 0.11 (P <0.05). The expressions of LGR5, SALL4 and NKD1 protein in lymph node metastasis group were 0.73 ± 0.11,0.74 ± 0.08 and 0.40 ± 0.11 respectively (P <0.05), and the expression of LGR5, SALL4 and NKD1 were 0.58 ± 0.13, 0.64 ± 0.14 and 0.57 ± 0.16 (P <0.05). LGR5, SALL4, NKD1 protein and breast cancer patients age, tumor size, tumor site has nothing to do (P> 0.05). Conclusion The abnormal expression of LGR5, SALL4 and NKD1 is closely related to the occurrence, development, invasion and metastasis of breast cancer. The combined detection of LGR5, SALL4 and NKD1 may play an important role in assessing the biological behavior of breast cancer. LGR5, SALL4 and NKD1 may lead to the occurrence and development of breast cancer.
