本文试验了低温后处理及其配合因素(2,4-D)对于小麦花药愈伤组织诱导的影响.结果表明,低温对提高愈伤组织诱导的影响是明显的,处理时间以2—3天为宜.低温的作用可能是以提高花粉细胞的存活率、延缓花粉正常发育、促进大多数花粉发育“同步化”等来影响愈伤组织的形成的.在低温后处理时,加入2,4-D与否,明显影响低温处理效果.对药壁组织较嫩材料,低温后处理效果以无2,4-D存在较好;药壁较老的材料则相反.2,4-D的作用可能也影响花粉的发育,低温和2,4-D的配合,可能既促进花粉细胞发育“同步化”,也有利于花粉发育和药壁发育的协调,从而影响愈伤组织的形成.因此,在低温处理的同时配合其他因素处理,可能为进一步弄清低温的作用机理,研究药壁的作用等,提供有用的线索.
