目的:研究NF-κB、PPAR-γ在Barrett食管及食管腺癌中的表达,探讨Barrett食管及食管腺癌的发病机制。方法:采用免疫组化染色检测正常食管上皮、Barrett食管黏膜及食管腺癌标本中NF-κB、PPAR-γ的蛋白表达情况。结果:NF-κB蛋白阳性染色位于细胞质,PPAR-γ蛋白阳性染色位于细胞核,NF-κB蛋白和PPAR-γ蛋白在正常鳞状上皮中均几乎不表达,在BE中呈轻中度表达,表达于杯状细胞及浅层腺体上皮,但在食管腺癌中广泛呈中强表达,与正常食管及BE比较,差异均有显著性(P<0.01),且在正常食管粘膜、BE及食管腺癌中的表达呈现逐渐升高的趋势。食管腺癌细胞分化程度和肿瘤病理学分期与NF-κB表达相关(P<0.05);癌细胞分化程度与PPAR-γ的表达相关(P<0.05)。结论:NF-κB与PPAR-γ在BE及食管腺癌中表达升高,说明两者的表达上调在BE及食管腺癌的发生发展中起了重要作用。
Objective: To investigate the expression of NF-κB and PPAR-γ in Barrett’s esophagus and esophageal adenocarcinoma and to explore the pathogenesis of Barrett’s esophagus and esophageal adenocarcinoma. Methods: Immunohistochemical staining was used to detect the protein expression of NF-κB and PPAR-γ in normal esophageal mucosa, esophageal mucosa of Barrett and esophageal adenocarcinoma. Results: The positive staining of NF-κB protein was located in the cytoplasm. The positive staining of PPAR-γ protein was located in the nucleus. The expression of NF-κB protein and PPAR-γ protein were almost not observed in normal squamous epithelium, but moderately and moderately expressed in BE In goblet cells and superficial gland epithelium, but in esophageal adenocarcinoma showed a strong moderate expression, compared with the normal esophagus and BE, the difference was significant (P <0.01), and in normal esophageal mucosa, BE and esophagus The expression of adenocarcinoma showed a gradual upward trend. The degree of esophageal adenocarcinoma differentiation and tumor pathological staging correlated with the expression of NF-κB (P <0.05). The differentiation of cancer cells was correlated with the expression of PPAR-γ (P <0.05). Conclusion: The expressions of NF-κB and PPAR-γ are up-regulated in BE and esophageal adenocarcinomas, indicating that the up-regulation of NF-κB and PPAR-γ may play an important role in the development of BE and esophageal adenocarcinoma.
