目的分析抗炎抗凝双效融合蛋白TAP-SSL5对动物的药理作用,为其安全性用药提供实验依据。方法取昆明(KM)小鼠,经尾静脉分别注射1、3和9 mg/kg TAP-SSL5,观察其对小鼠神经系统的影响;取SD大鼠,经尾静脉分别注射1和3 mg/kg TAP-SSL5,设溶剂[20 mmol/L甘氨酸(Gly)-NaOH缓冲液,pH 10.6]为对照组,观察其对呼吸系统和心血管系统的影响。结果 TAP-SSL5对KM小鼠一般行为状态、协调机能和戊巴比妥钠阈下剂量催眠的协同作用无明显影响;对SD大鼠的血压、呼吸频率和心率无明显影响,与溶剂对照组比较,差异无统计学意义(P均>0.05)。结论融合蛋白TAP-SSL5对动物的神经系统、呼吸系统和心血管系统无明显影响,表明其具有较好的安全性,且无明显的毒副作用。
Objective To analyze the pharmacological effects of anti-inflammatory and anticoagulant double-acting fusion protein TAP-SSL5 on animals and provide experimental evidence for its safety. Methods Kunming (KM) mice were injected with 1, 3 and 9 mg / kg TAP-SSL5 via the tail vein respectively to observe their effects on the nervous system. SD rats were injected with 1 and 3 mg / kg TAP-SSL5, and the solvent [20 mmol / L Gly-NaOH buffer pH 10.6] as the control group to observe the respiratory and cardiovascular system. Results TAP-SSL5 had no significant effect on the general behavioral status, coordination function and hypnotic synergy of sublingual dose of pentobarbital sodium in KM mice, but no significant effect on blood pressure, respiratory rate and heart rate in KM rats. Compared with the solvent control group The difference was not statistically significant (all P> 0.05). Conclusion The fusion protein TAP-SSL5 has no significant effect on the nervous system, respiratory system and cardiovascular system of animals, indicating that it has good safety and no obvious side effects.
