目的比较干扰素α-1b(IFNα-1b)与聚乙二醇化干扰素α-1b(PEGIFNα-1b)抑制荷HepG-2裸鼠肿瘤形成的效果。方法取对数生长期的人肝癌HepG-2细胞接种于BALB/c雄性裸鼠背部皮下制备荷瘤动物模型。接种后14d按肿瘤大小将荷HepG-2瘤鼠随机分为溶剂对照组、PEGIFNα-1b组和IFNα-1b组3组,均连续5周皮下注射,每周1次。用电子数显卡尺测量肿瘤体积,第5周末全身麻醉处死裸鼠后测量体重、肿瘤体积和重量。结果观察期内PEGIFNα-1b和IFNα-1b对肿瘤的抑制率分别达56%和42%,未见其对荷瘤鼠体重有明显影响。结论PEGIFNα-1b对裸鼠体内HepG-2肿瘤生长有明显的抑制作用。
Objective To compare the inhibitory effects of interferon α-1b (IFNα-1b) and pegylated interferon α-1b (PEGIFNα-1b) on the tumor formation in HepG-2-bearing nude mice. Methods Human hepatoma HepG-2 cells in logarithmic growth phase were inoculated subcutaneously into BALB / c male nude mice to prepare tumor-bearing animal model. On the 14th day after inoculation, HepG-2 tumor-bearing mice were randomly divided into solvent control group, PEGIFNα-1b group and IFNα-1b group according to tumor size. All mice were injected subcutaneously once weekly for 5 weeks. Tumor volume was measured with an electronic digital caliper. Body weight, tumor volume and weight were measured after the nude mice were sacrificed by general anesthesia on the fifth weekend. Results The inhibitory rates of PEGIFNα-1b and IFNα-1b on tumor were 56% and 42% respectively during the observation period, and no significant effect on body weight of tumor-bearing mice was observed. Conclusion PEGIFNα-1b significantly inhibits HepG-2 tumor growth in nude mice.
