目的采用TUNEL技术对福尔马林固定、石蜡包埋的肝组织和培养肝癌细胞进行染色,以探讨肝细胞凋亡的形态特征和分布.方法使用德国宝灵曼公司生产的原位末端标记试剂盒.石蜡包埋组织经微波处理后,行常规TUNEL染色,观察凋亡细胞的形态和分布.结果TUNEL阳性信号为黄绿色或黄色荧光,位于胞核,呈小圆形或环行.TUNEL阳性细胞大多在形态学上表现为凋亡的细胞.对肝组织TUNELLI的分析显示,肝癌组织中细胞凋亡指数(0395)比肝硬变(0075)和癌旁肝组织(0105)要高,且癌组织恶性程度越高,这种凋亡就越显著.结论改进的TUNEL技术对福尔马林固定、石蜡包埋的难处理组织进行染色是一个有效的方法.
Objective To dye formalin-fixed, paraffin-embedded liver tissue and cultured hepatoma cells by TUNEL technique to investigate the morphological characteristics and distribution of hepatocyte apoptosis. Methods The in situ end-labeling kit produced by German Bolinger Mann was used. After paraffin-embedded tissue was treated with microwave, normal TUNEL staining was performed to observe the morphology and distribution of apoptotic cells. Results The TUNEL positive signal was yellow-green or yellow fluorescence, located in the nucleus, showing small round or circular lines. Most of the TUNEL positive cells were morphologically apoptotic cells. Analysis of TUNELLI in liver tissue showed that the apoptosis index (0395) was higher in liver cancer tissue than in liver cirrhosis (0075) and para-carcinoma tissue (0105), and the degree of malignancy of cancer tissue was higher. The more significant this apoptosis is. 5. Conclusion The improved TUNEL technique is an effective method for staining formalin-fixed, paraffin-embedded, intractable tissues.
