目的研究结核病患者巨噬细胞移动抑制因子(migration inhibitory factor,MIF)表达水平,分析MIF-173G/C多态性与结核杆菌易感性关系。方法选取第三军医大学大坪医院野战外科研究所呼吸科2009年9月至2010年9月结核病患者68例及70例健康体检者,采用RFLP-PCR的方法分析2组MIF-173基因型,ELISA法检测血清中MIF水平。结果结核病组中MIF-173 CC纯合基因型及MIF-173*C等位基因频率较正常对照组明显升高(OR值分别为3.99和1.58,P=0.003和0.02),结核组MIF水平明显高于对照组[(705.21±67.98)pg/mlvs(355.31±57.29)pg/ml,P<0.01],表达水平平均升高1倍左右。结论 MIF-173*C基因型与结核杆菌易感有关,在结核病发病中可能发挥重要作用。
Objective To study the expression level of MIF in M tuberculosis patients and to analyze the relationship between the MIF-173G / C polymorphism and susceptibility to Mycobacterium tuberculosis. Methods Totally 68 patients with tuberculosis and 70 healthy subjects from Department of Respiratory Diseases, Field Institute of Field Surgery, Daping Hospital, Third Military Medical University were enrolled. Two groups of MIF-173 genotypes were analyzed by RFLP-PCR. Method to detect serum MIF levels. Results The frequencies of MIF-173 CC homozygous MIF-173 * C allele and MIF-173 * C allele in patients with tuberculosis were significantly higher than those in controls (OR = 3.99 and 1.58, P = 0.003 and 0.02, respectively) Which was higher than that in the control group [(705.21 ± 67.98) pg / ml vs (355.31 ± 57.29) pg / ml, P <0.01]. Conclusion The MIF-173 * C genotype is associated with the susceptibility of Mycobacterium tuberculosis and may play an important role in the pathogenesis of tuberculosis.
