目的:比较固本健脑法、补脾健脑法、补肾健脑法对老年健忘大鼠海马组织中AGEs/RAGE信号通路的影响。方法:腹腔注射D-半乳糖及Na NO2建立老年健忘大鼠模型,分别灌胃固本健脑方、归脾丸、六味地黄丸和石杉碱甲;并设空白组和模型组,灌以生理盐水。给药结束后,取大鼠海马组织,采用免疫组化法检测AGEs的含量,Western blot检测RAGE、NF-κB p65的表达,ELISA检测IL-1β、TNF-α的含量。结果:与空白组比较,模型组、西药组、补脾组、补肾组、固本组上述指标的表达均上调;与模型组比较,西药组、补脾组、补肾组、固本组上述指标的表达均下调;与固本组比较,西药组、补脾组、补肾组上述指标的表达下调;补脾组、补肾组、西药组组间比较,差异无统计学意义。结论:固本健脑法、补脾健脑法、补肾健脑法均能在一定程度上改善老年健忘大鼠的记忆功能,但固本健脑法作用最佳,固本健脑法改善记忆功能的分子机制与AGEs/RAGE信号通路密切相关。
Objective: To compare the effects of Guben Jiannao Decoction, BuBu Jiannao Decoction, and BushennaoJiannao Decoction on the AGEs / RAGE signal pathway in hippocampus of aged rats. Methods: The aged amnesia rats model was established by intraperitoneal injection of D-galactose and NaNO2. Gubengjiannao, Guipi Wan, Liuweidihuang Pills and huperzine A were given respectively. The rats in the blank group and the model group were given physiological saline brine. After the administration, the hippocampus tissues of rats were taken out. The contents of AGEs were detected by immunohistochemistry. The expressions of RAGE and NF-κB p65 were detected by Western blot. The levels of IL-1β and TNF-α were detected by ELISA. Results: Compared with the blank group, the expression of the above indexes were up-regulated in the model group, the western medicine group, the spleen-supplementing group, the kidney-invigorating group and the solid-root group. Compared with the model group, The expression of the above indexes was down-regulated in the western medicine group, the spleen-supplementing group and the kidney-invigorating group compared with the control group. There was no significant difference between the two groups. Conclusion: Gubenjiannaotan, Bupijiannao, Bushenjiannao can all improve the memory function of senile amnesia rats to a certain extent, but Gubenjiannao method has the best effect, Gubenjiannao method to improve memory The molecular mechanism of function is closely related to the AGEs / RAGE signaling pathway.
