利用高效液相色谱-蒸发光散射检测器定量检测啤酒中的多糖化合物,并对方法进行优化和验证。检测条件如下:5μm NH_2色谱柱;乙腈/水梯度洗脱;各糖组分浓度的线性范围为,果糖:0.05-5.00g/L,葡萄糖:0.05-5.00g/L,麦芽糖:0.05-15.00g/L,麦芽三糖:0.05-10.00g/L,麦芽四糖:0.05-5.00g/L,上述各糖组分的检测限分别为0.005g/L、0.008g/L、0.01g/L、0.01g/L、0.01g/L。该检测方法的精密度和精确度在低酒精度、6%酒精度和添加辅助物进行发酵(4.5%酒精度)三种酒体中进行验证。相对标准偏差在1.59-5.95%范围内,回收率在94-98 4%范围内。
The polysaccharide compounds in beer were quantified by HPLC - Evaporative light scattering detector, and the method was optimized and validated. The detection conditions were as follows: 5μm NH_2 column; acetonitrile / water gradient elution; the linear range of each sugar component concentration was fructose 0.05-5.00g / L, glucose 0.05-5.00g / L, maltose 0.05-15.00g / L, maltotriose 0.05-10.00g / L, and maltotetraose 0.05-5.00g / L. The detection limits of the above sugar components are respectively 0.005g / L, 0.008g / L and 0.01g / L, 0.01g / L, 0.01g / L. The precision and precision of the assay was verified in three bod- ows including low alcohol, 6% alcohol and fermented (4.5% alcohol) supplements. The relative standard deviations were in the range of 1.59-5.95% and the recoveries were in the range of 94-98 4%.
