目的观察自发性高血压大鼠(SHR)肾小球及肾小管间质CD2相关蛋白(CD2AP)水平和肾脏纤维化之间的关系。方法22只16周龄雄性SHR随机分成干预组和对照组,每组各11只,干预组给予依普利酮(每日100mg/kg)。观察10周,检测血压、血清肌酐(Cr)、尿素氮(BUN)、尿酸(UA)、肾组织CD2AP和转化生长因子β1(TGF-β1)蛋白水平。行肾组织HE和Masson染色,检测病理切片并进行评分。结果对照组血压高于干预组(P<0.01),干预组肾小球胶原纤维含量和肾小管间质评分低于对照组(P<0.05),干预组TGF-β1和CD2AP蛋白水平低于对照组(P<0.01)。大鼠肾小球与肾小管间质CD2AP蛋白水平和BUN、Cr、UA无相关性(P>0.05),和相应的TGF-β1蛋白水平、肾小球胶原纤维含量与肾小管间质评分呈负相关(r值分别为-0.894,-0.921,-0.798和-0.532,P<0.01)。结论16周龄SHR肾小球和肾小管间质CD2AP蛋白水平降低,可能导致肾脏纤维化。
Objective To observe the relationship between CD2AP and renal fibrosis in glomeruli and tubulointerstitium of spontaneously hypertensive rats (SHRs). Methods Twenty-two male SHRs of 16 weeks old were randomly divided into intervention group and control group, with 11 rats in each group. Eplerenone (100 mg / kg daily) was administered to the intervention group. After 10 weeks of observation, blood pressure, serum creatinine (Cr), blood urea nitrogen (BUN), uric acid (UA), renal tissue CD2AP and TGF-β1 protein levels were measured. The renal tissue was stained with HE and Masson, the pathological sections were taken and scored. Results The blood pressure of the control group was higher than that of the intervention group (P <0.01). The contents of glomerular collagen fibers and tubulointerstitial were lower in the intervention group than those in the control group (P <0.05). The levels of TGF-β1 and CD2AP in the intervention group were lower than those in the control group Group (P <0.01). There was no correlation between CD2AP level and BUN, Cr and UA in glomeruli and tubulointerstitium (P> 0.05), and the corresponding TGF-β1 protein level, glomerular collagen fibers and tubulointerstitial score (R values were -0.894, -0.921, -0.798 and -0.532 respectively, P <0.01). Conclusion The level of CD2AP protein in glomeruli and tubulointerstitial tissue of 16-week-old SHRs may be decreased, which may lead to renal fibrosis.
