目的:通过测定冷冻(T=-18℃)、冷藏(T=4℃)保存条件下环孢菌素A全血样品的浓度,研究环孢菌素A的稳定性。方法:本实验采用荧光偏振免疫法(FPIA)测定环孢菌素A的浓度。结果:冷藏保存20天、冷冻保存30天后的样品浓度与取血当日的样品浓度有统计学差异。结论:环孢菌素A的全血样本冷冻保存少于20天,冷藏保存少于15天测得的浓度与取血当日测得的浓度无统计学差异,因此环孢菌素A全血样本可以在冷藏条件下保存15天,在冷冻条件下保存20天。
OBJECTIVE: To study the stability of cyclosporin A by measuring the concentration of cyclosporine A whole blood sample under the conditions of freezing (T = -18 ℃) and refrigeration (T = 4 ℃). Methods: In this experiment, the concentration of cyclosporin A was determined by fluorescence polarization immunoassay (FPIA). Results: There was a significant difference between the samples stored in refrigerator for 20 days after cryopreservation for 30 days and the samples taken on the day of blood sampling. CONCLUSIONS: Cyclosporin A whole blood samples are stored frozen for less than 20 days. There is no significant difference between the concentrations measured after refrigerated storage for less than 15 days and those measured on the day of blood sampling. Therefore, the cyclosporin A whole blood sample It can be stored for 15 days under refrigeration and 20 days under frozen conditions.
