目的:研究葛根素对同型半胱氨酸(homocysteinemia,HCY)诱导增殖的血管平滑肌细胞(vascular smooth musclecell,VSMC)细胞周期的影响及其机制。方法:建立HCY诱导的兔VSMC增殖模型,予不同浓度的葛根素(10~(-7) mol/L,10~(-6)mol/L,10~(-5)mol/L)共同培养48h后,用MTT法检测细胞增殖能力;用流式细胞仪Annexin/PI双染法检测细胞周期变化及细胞周期调控相关蛋白CyclinD1和CDK4的表达。结果:葛根素可抑制HCY所诱导的VSMC增殖,抑制作用与葛根素浓度呈正相关;葛根素组G_0/G_1期细胞数明显增多,S期细胞数明显减少,与HCY模型组比较,差异均有非常显著性意义(P<0.01);CyclinD1和CDK4的表达量均随葛根素浓度增加而减少。结论:葛根素拮抗HCY诱导的兔VSMC增殖,主要机制之一可能与CyclinD1及CDK4低表达所致的细胞周期抑制有关。
Objective: To investigate the effect of puerarin on the cell cycle of vascular smooth muscle cells (VSMCs) induced by homocysteinemia (HCY) and its mechanism. Methods: HCY-induced rabbit VSMC proliferation model was established and co-cultured with different concentrations of puerarin (10 -7 mol / L, 10 -6 mol / L and 10 -5 mol / L) After 48h, the cell proliferation was detected by MTT assay. The cell cycle and the expression of CyclinD1 and CDK4 were detected by flow cytometry Annexin / PI double staining. Results: Puerarin inhibited the proliferation of VSMC induced by HCY, and the inhibition was positively correlated with the concentration of puerarin. The number of cells in G0 / G1 phase of puerarin increased significantly and the number of cells in S phase decreased significantly. Compared with HCY model group, (P <0.01). The expression of CyclinD1 and CDK4 decreased with the increase of puerarin concentration. CONCLUSION: Puerarin can antagonize the proliferation of VSMC induced by HCY. One of the mechanisms may be related to the inhibition of cell cycle induced by low expression of CyclinD1 and CDK4.
