目的测定胡芦巴树脂提取物中总黄酮和牡荆素的含量。方法以牡荆素为对照品,采用UV法于340nm处测定总黄酮;采用HPLC法测定牡荆素,C18色谱柱(150mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸水溶液(16:84),检测波长340nm。结果总黄酮3.18~19.08μg·mL-1(r=0.9998)、牡荆素0.015~0.241μg(r=0.9998)线性关系良好;总黄酮和牡荆素的平均回收率分别为99.3%、100.47%;3批样品中总黄酮含量为16.24%、16.90%、15.95%,牡荆素含量为2.27%、2.42%、2.26%。结论所建方法简便、快速、准确,适用于胡芦巴树脂提取物中总黄酮和牡荆素的测定。
Objective To determine the content of total flavonoids and vitexin in fenugreek resin extract. Methods Vitexin was used as a reference substance and the total flavonoids were determined by UV method at 340 nm. The vitexin and C18 column (150 mm × 4.6 mm, 5 μm) were determined by HPLC. The mobile phase was acetonitrile-0.1% phosphoric acid solution (16 : 84), detection wavelength 340nm. The results showed that the average recoveries of total flavonoids and vitexin were 99.3% and 100.47%, respectively. The average recoveries of total flavonoids and vitexin were 3.18-19.08 μg · mL-1 (r = 0.9998) and 0.015-0.241 μg (r = 0.9998) The content of total flavonoids in three batches of samples was 16.24%, 16.90% and 15.95%, and the content of vitexin was 2.27%, 2.42% and 2.26% respectively. Conclusion The method is simple, rapid and accurate and is suitable for the determination of total flavonoids and vitexin in fenugreek resin extract.
