运用花粉管导入和农杆菌介导两种方法,对泡桐转TCS基因受体表现性状进行研究,结果表明:泡桐花导入TCS基因后导致泡桐坐果率和畸形率升高;农杆菌介导TCS基因于泡桐组培苗的愈伤组织、茎段、茎尖后,致使其死亡率、分化率、分化数、苗高和颜色发生显著变化,极差分析表明,最佳转基因方式是以茎尖为受体,预培养3d,浸染时间15min,共培养条件为半黑暗。
Pollen tube induction and Agrobacterium-mediated two kinds of methods, the Paulownia transferred TCS gene receptor traits were studied, the results show that: Paulownia flower into the TCS gene leads to Paulownia fruit set and the rate of abnormalities; Agrobacterium-mediated TCS gene The results showed that the mortality, differentiation rate, differentiation number, seedling height and color of Callus paeoniflorum were significantly changed after the callus, stem segment and shoot tip of the tissue culture seedling. The difference analysis showed that the best transgenic method was that the shoot tip was Receptor, pre-culture 3d, dipping time 15min, co-culture conditions were semi-dark.
