目的观察昆母汤醇提液对人类风湿关节炎(RA)成纤维样滑膜细胞(FLS)凋亡及对caspase-3表达的影响,探讨昆母汤治疗RA的部分作用机制。方法滑膜组织取自行关节置换术或关节镜检查术的8例活动性RA患者,分离出FLS进行原代培养,采用流式细胞仪检测法检测昆母汤醇提液对RA-FLS细胞凋亡率的影响,采用酶联免疫吸附测定法检测培养上清液caspase-3蛋白含量的变化。结果昆母汤醇提液和甲氨蝶呤均可显著促进RA-FLS的凋亡,且昆母汤醇提液促RA-FLS凋亡能力在一定程度上呈剂量依赖性。高剂量昆母汤醇提液可使TNF-α诱导的caspase-3表达量增加。结论昆母汤可能通过促进caspase-3的表达来促进RAFLS凋亡,具有治疗RA的潜在价值。
Objective To observe the effect of Kunmu decoction on the apoptosis of rheumatoid arthritis (RA) fibroblast-like synoviocytes (FLS) and the expression of caspase-3 in human rheumatoid arthritis (RA) and to explore the mechanism of Kunmu decoction in treating RA. Methods Eight patients with active RA who underwent arthroplasty or arthroscopy were treated with synovial membrane. FLS was isolated and cultured in primary culture. Flow cytometry was used to detect the effect of Kunmutang alcohol extract on RA-FLS cells Apoptosis rate, the use of enzyme-linked immunosorbent assay to detect the culture supernatant caspase-3 protein content changes. Results Both Kunmutang alcohol extract and methotrexate could significantly promote the apoptosis of RA-FLS, and the Kun-Tang alcohol extract promoted the apoptosis of RA-FLS to a certain extent in a dose-dependent manner. High-dose Kun-Tang alcohol extract can increase TNF-α-induced caspase-3 expression. Conclusions Kunmu Decoction may promote the apoptosis of RAFLS by promoting the expression of caspase-3, which has the potential value of treating RA.
