目的研究鼠Leydig细胞膜LH受体减量调节中受体mRNA及cAMP水平的变化及二者之间的关系。方法应用RT-PCR法,以β-actin为内参照,测定激素刺激后鼠Leydig细胞膜LH受体(LHR)减量调节中LHRmRNA水平的变化,同时应用放射免疫法,分析同一时间细胞内cAMP水平的变化。结果LHRmRNA水平早期有20%的升高,然后逐渐下降50%,与此趋势相同,cAMP水平亦呈现出升高-降低的动态过程,二者在变化上呈“正相关”,并且前者总是“滞后”于后者一段时间。结论LHR减量调节过程中,细胞内cAMP水平可能以“正相关”的方式影响着LHR基因的转录速率。
Aim To investigate the changes of receptor mRNA and cAMP in LH receptor of Leydig cell membrane and the relationship between them. Methods RT-PCR and β-actin were used as internal reference to measure LHR mRNA level in LHR of LHR (Leydig’s cell membrane) after stimulated by hormone. Radiofrequencyimmunoassay was used to analyze intracellular cAMP level at the same time The change. Results The level of LHR mRNA increased by 20% in the early stage and then gradually decreased by 50%. With the same trend, the level of cAMP also showed a dynamic process of increase-decrease. Both of them showed “positive correlation” with the changes, and the former always The “lag” in the latter period of time. Conclusion During the process of LHR decrement, the level of intracellular cAMP may affect the transcription rate of LHR gene in a “positive correlation” manner.
