目的观察肾衰养真胶囊对腺嘌呤致慢性肾衰竭(CRF)营养不良大鼠下丘脑瘦素(Leptin)及其长型受体(OB-Rb)基因表达的影响。方法采用0.5%腺嘌呤+4%酪蛋白饲料喂养制作CRF营养不良大鼠模型,观察其营养不良发生时间,符合CRF营养不良模型的大鼠随机分为模型组、肾衰养真胶囊组和开同组,另取正常大鼠为正常对照组。灌胃4周,检测血肌酐(Scr)、尿素氮(BUN)、24 h尿蛋白定量(24 hUpr)、白蛋白(Alb)、血红蛋白(Hb),采用放免法检测血清Leptin水平,RT-PCR法检测下丘脑OB-Rb mRNA表达。结果大鼠在造模8周末出现营养不良,肾衰养真胶囊可增加Alb、Hb水平,降低Scr、BUN、24 hUpr水平,降低血清Leptin水平,下调下丘脑OB-RbmRNA表达。结论肾衰养真胶囊可通过降低血清Leptin水平,下调下丘脑OB-Rb mRNA表达,从而改善CRF营养不良。
Objective To observe the effect of Shenshuaiyangzhen capsule on hypothalamic leptin and its long receptor (OB-Rb) gene expression in adenine-induced chronic renal failure (CRF) malnourished rats. Methods The CRF malnutrition rat model was fed with 0.5% adenine + 4% casein feed to observe the time of malnutrition. The rats that matched the CRF malnutrition model were randomly divided into model group, Shenshiai Yangzheng capsule group and open group. In the same group, another normal rat was used as a normal control group. After intragastric administration for 4 weeks, serum creatinine (Scr), urea nitrogen (BUN), 24-hour urinary protein (24 hUpr), albumin (Alb), and hemoglobin (Hb) were measured. Serum leptin levels were measured by radioimmunoassay, and RT-PCR was performed. The hypothalamus OB-Rb mRNA expression was measured by the method. Results The rats suffered from malnutrition at the end of the 8th week of modeling. Shenzhi Yangzheng Capsules increased Alb and Hb levels, decreased Scr, BUN, and 24 hUp levels, decreased serum leptin levels, and down-regulated the expression of OB-Rb mRNA in the hypothalamus. Conclusion Shenshuaiyangzhen capsule can improve the malnutrition of CRF by decreasing serum leptin levels and down-regulating OB-Rb mRNA expression in hypothalamus.
