(目的〕研究肺癌中MTS1/p16和p53基因产物的表达与细胞增殖的关系。〔方法〕应用SP免疫组织化学方法研究62例肺癌组织中p16蛋白和p53蛋白的表达情况,并进行增殖细胞核抗原检测,计算细胞增殖指数(PI)。(结果)62例肺癌组织中p16蛋白和p53蛋白阳性率分别为58.1%和59、7%。晚癌p16蛋白的阳性率明显高于小细胞癌(p<0.05);淋巴结转移阳性组p16蛋白的表达显著低于阴性组(P<0.05);PI分级为巨级的p16蛋白表达显著高于Ⅳ级(p<0.05)。不同组织类型肺癌中p53蛋白的表达未见明显差异,淋巴结转移阳性组p53蛋白的表达高于阴性组(p<0.01〕;不同PI分级中p53蛋白的表达,N级明显高于1级和Ⅱ级(P<0.05),三级明显高于1级(p<0.05)和Ⅱ级(P<0.01)。p16蛋白低表达和p53蛋白过度表达之间未见明显相关性。(结论〕提示p16蛋白低表达和p53蛋白过度表达均有促进肺癌细胞增殖的作用,p16蛋白的表达与肺癌的细胞分化有关,p53蛋白过度表达对肺癌细胞的转移起重要作用。抑癌基因p53对MTS1/p16基因无明显调控作用。检测p53蛋白表达可作为肺癌诊断的一项新指标。
(Objective) To study the relationship between the expression of MTS1/p16 and p53 gene products and cell proliferation in lung cancer.Methods SP immunohistochemical method was used to study the expression of p16 protein and p53 protein in 62 cases of lung cancer, and to perform proliferating cell nuclear antigen. Detection and calculation of cell proliferation index (PI) (Results) The positive rates of p16 protein and p53 protein in 62 lung cancer tissues were 58.1% and 59, 7%, respectively.The positive rate of p16 protein in late cancer was significantly higher than that of small cell carcinoma. (p<0.05); The expression of p16 protein was significantly lower in the positive lymph node metastasis group than in the negative group (P<0.05); the expression of the p16 protein in the PI grade was significantly higher than that in the IV grade (p<0.05). There was no significant difference in the expression of p53 protein in lung cancer. The expression of p53 protein in lymph node metastasis positive group was higher than that in negative group (p<0.01). The expression of p53 protein in different PI grades was significantly higher than that in grade 1 and grade II (P). <0.05), Grade 3 was significantly higher than grade 1 (p<0.05) and grade II (P<0.01).There was no significant correlation between p16 protein low expression and p53 protein overexpression. Low expression of p16 protein and p53 protein The expression of p16 protein is related to the cell differentiation of lung cancer, and the overexpression of p53 protein plays an important role in the metastasis of lung cancer cells.The tumor suppressor gene p53 has no obvious regulation on MTS1/p16 gene. Protein expression can be used as a new indicator of lung cancer diagnosis.
