目的通过体外实验研究Her-2阳性胃癌细胞株对曲妥珠单抗的药物敏感性。方法采用Western-blot法检测人胃癌细胞系NCI-N87、SGC7901、MKN48以及MKN25等4种胃癌细胞株的Her-2蛋白表达水平;采用MTT法检测曲妥珠单抗对细胞株的抑制率。结果 4种胃癌细胞株均存在Her-2蛋白表达,其中NCI-N87细胞表达水平最高(P=0.000);曲妥珠单抗浓度分别为10μg/ml、20μg/ml以及40μg/ml时,NCI-N87细胞抑制率最高(均P=0.000),半数抑制浓度IC50最低;随着曲妥珠单抗浓度的增加,4种胃癌细胞株的细胞抑制率均增加。结论曲妥珠单抗能抑制Her-2阳性胃癌细胞株的生长,这种抑制作用与细胞Her-2蛋白表达水平相关,并且存在剂量依赖性。
Objective To study the drug sensitivity of Her-2 positive gastric cancer cell lines to trastuzumab in vitro. Methods Western-blot was used to detect Her-2 protein expression in human gastric cancer cell lines NCI-N87, SGC7901, MKN48 and MKN25. The inhibition rate of trastuzumab on cell lines was detected by MTT assay. Results The expression of Her-2 protein was found in all the 4 gastric cancer cell lines, among which the expression level of NCI-N87 cells was the highest (P=0.000); the concentrations of trastuzumab were 10 μg/ml, 20 μg/ml and 40 μg/ml, respectively. The inhibition rate of N87 cells was the highest (all P=0.000), and the IC50 of half inhibition was the lowest. With the increase of trastuzumab concentration, the inhibition rates of the four gastric cancer cell lines increased. Conclusion Trastuzumab can inhibit the growth of Her-2 positive gastric cancer cell lines, and this inhibition is related to the expression level of Her-2 protein in a dose-dependent manner.
