为观察IL 1β预处理后心肌细胞粘着分子(CAMs)表达的变化及其与延迟保护 (DP)作用的关系 ,在大鼠心肌缺血再灌注 (I/R)模型上设假手术对照组(C组 )、生理盐水组 (NS组 )和IL 1β预处理组 (ILPC组 ) ,采用原位杂交、免疫组化及酶法检测低剂量IL 1β注入大鼠后即刻、12h和 2 4h心肌细胞间粘附分子 1(ICAM 1)mRNA及其蛋白质表达和白细胞功能相关抗原 1(CD11a)蛋白质表达的变化 ,以及中性粒细胞(PMNs)的浸润数 ,并测定心肌梗死面积。结果发现 ,与C组比较 ,NS组和ILPC组各时相点ICAM 1mRNA及其蛋白质表达和CD11a蛋白质表达以及PMNs浸润数均显著增加 (P <0 0 5或 0 0 1) ,但ILPC组后 2 4hI/R末 ,上述指标的增幅明显减少 (P <0 0 5 ) ,且心肌梗死面积显著下降 (P <0 0 5 )。即刻和 12h,ILPC组与NS组比较差异无显著性意义 (P >0 0 5 )。提示 ,低剂量IL 1β预处理可使心肌延迟期CAMs表达减少而产生延迟保护作用
To observe the changes of the expression of adhesion molecules (CAMs) in cardiomyocytes after IL 1β preconditioning and its relationship with delayed protection (DP), a sham operation control group (I / R) (Group C), saline group (NS) and IL 1β preconditioning group (ILPC group). Compared with group C, ICAM 1 mRNA and protein expression, CD11a protein expression and PMNs infiltration in NS group and ILPC group at each time point were significantly increased (P <0.05 or 0.01) At 24 h I / R, the increase of these indexes was significantly decreased (P <0.05), and the area of myocardial infarction was significantly decreased (P <0.05).
