生长素酰胺水解酶(IAA-Leucine Resistant1-like Hydrolase,ILL)可催化生长素结合态释放游离态生长素(IAA)。为了深入研究桃ILL基因家族的功能,对桃基因组中ILL基因家族成员的数量、在染色体骨架的分布、编码蛋白质的结构、基因表达模式和编码氨基酸的进化树分类等方面进行了研究。桃基因组中共鉴定出了9个ILL基因,它们分布在4个染色体骨架上(scaffold 1、2、3和7)。在桃中编码ILL蛋白家族的基因成员分别为:Pp ILR1、Pp ILR2、Pp ILR3、Pp ILR4、Pp ILL1、Pp ILL2、Pp ILL3、Pp ILL4和Pp ILL5,所有这些预测的蛋白都含有M20结构域和M20二聚化结构域。运用荧光定量实时PCR技术对该基因家族在桃果实生长发育各阶段的表达水平进行了分析,结果显示:ILL基因家族中的数个成员在果实不同发育阶段表达水平有变化,其中Pp ILR1的表达量在果实生理成熟期明显上调。
IAA-Leucine Resistant1-like Hydrolase (ILL) catalyzes the release of free auxin (IAA) by auxin-bound states. In order to further study the function of peach ILL gene family, the number of ILL gene family members in peach genome, the distribution of the chromosomal skeleton, the structure of the encoded protein, the pattern of gene expression and the classification of amino acid-encoding phylogenetic trees were studied. Nine ILL genes were identified in the peach genome and distributed over four chromosomal backbones (scaffolds 1, 2, 3 and 7). The gene members that encode the ILL protein family in peach are: Pp ILR1, Pp ILR2, Pp ILR3, Pp ILR4, Pp ILL1, Pp ILL2, Pp ILL3, Pp ILL4 and Pp ILL5, all of these predicted proteins contain the M20 domain And M20 dimerization domains. Fluorescence quantitative real-time PCR was used to analyze the expression level of this gene family at different stages of peach fruit growth and development. The results showed that several members of ILL gene family had different expression levels at different developmental stages of fruit, among which the expression of Pp ILR1 The amount of fruit in the physiological maturity significantly increased.
