目的 以LacZ基因为报告基因,建立新的大鼠脑胶质瘤动物模型。方法用BAG质粒转染C6胶质瘤细胞,体外观察并比较了野生型和转基因瘤细胞的生长周期、细胞增殖情况和恶性侵袭能力,利用立体定向技术建立了大鼠颅内外的肿瘤实验动物模型。结果 基因转染瘤细胞和野生型瘤细胞的生长特性和致瘤能力无明显差别。结论 以转基因瘤细胞建立的动物模型,能从细胞水平观察到胶质瘤的侵袭范围,为进一步进行脑胶质瘤的放疗和化疗实验,提供了一个敏感动物模型。
Objective LacZ gene as a reporter gene to establish a new animal model of rat glioma. Methods C6 glioma cells were transfected with BAG plasmid. The growth cycle, cell proliferation and malignant invasiveness of wild-type and transgenic tumor cells were observed and compared in vitro. The experimental animal models of intracranial and extracranial tumors were established by stereotactic technique . Results The growth characteristics and tumorigenicity of gene-transfected and wild-type tumor cells showed no significant difference. Conclusion The animal model established by transgenic tumor cells can observe the extent of glioma invasion from the cellular level and provide a sensitive animal model for radiotherapy and chemotherapy of glioma.
