目的研究野西瓜挥发油的抗肿瘤作用。方法采用MTT实验和SRB实验观察野西瓜挥发油对SGC-7901细胞的体外抗肿瘤作用;采用倒置荧光显微镜观察野西瓜挥发油作用后典型的细胞凋亡形态;采用流式细胞仪测定野西瓜挥发油对SGC-7901细胞凋亡及细胞周期的影响。结果10~200μg/mL的野西瓜挥发油作用于SGC-7901细胞72h后均可不同程度的抑制SGC-7901细胞的增殖,其IC50为145.5μg/mL,GI50为121.32μg/mL,LC50为900.96μg/mL,TGI为240.55μg/mL;作用24h后出现典型的凋亡形态;75、150、300μg/mL的野西瓜挥发油作用24h时出现细胞凋亡;周期分布发生改变,呈现S期阻滞。结论野西瓜挥发油具有明显抑制SGC-7901细胞增殖作用,其SGC-7901机制为诱导肿瘤细胞凋亡及改变细胞周期。
Objective To study the antitumor effect of the volatile oil of wild watermelon. Methods MTT assay and SRB assay were used to observe the anti-tumor effects of volatile oil of wild watermelon on SGC-7901 cells in vitro. Inverted fluorescence microscopy was used to observe the typical apoptotic morphology of the volatile oil of wild watermelon. Flow cytometry -7901 Cell Apoptosis and Cell Cycle. Results SGC-7901 cells were inhibited proliferation by SGC-7901 cells with 10 ~ 200μg / mL volatile oil for 72h. IC50 was 145.5μg / mL, GI50 was 121.32μg / mL, LC50 was 900.96μg / mL and TGI was 240.55μg / mL. Apoptotic morphology was observed after 24 hours of action. Apoptosis was observed at 24 hours after treatment with volatile oil of 75,150,300μg / mL. The cycle distribution changed and S phase arrest occurred. Conclusion Volatile oil from wild watermelon can significantly inhibit the proliferation of SGC-7901 cells. The mechanism of SGC-7901 is to induce tumor cell apoptosis and change the cell cycle.
