目的观察红三叶草异黄酮对非酒精性脂肪肝大鼠肝功能及炎症反应影响。方法雄性SD大鼠随机分为对照组(喂普通饲料)、造模组(脂肪乳灌胃),同时给予18%蔗糖溶液,连续4周,将造模成功大鼠按体重随机分为模型组、红三叶草低、中、高剂量组(75、150、300 mg/kg红三叶草异黄酮灌胃),连续12周,检测血清及肝组织中谷草转氨酶(AST)、谷丙转氨酶(ALT)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、C反应蛋白(CRP)的活性或含量。结果与模型组比较,红三叶草组肝细胞损伤减轻,肝功能改善,中剂量红三叶草组大鼠血清ALT[(65.63±13.00)U/L]、AST[(145.88±34.28)U/L]含量明显降低(P<0.05);中剂量红三叶草组大鼠血清TNF-α、IL-6、CRP分别为(43.24±4.14)ng/mL、(69.94±6.98)pg/mL、(3.06±0.36)mg/L,均明显低于模型组(P<0.05)。结论红三叶草异黄酮对非酒精性脂肪肝大鼠的肝功能具有保护作用,可降低炎症反应。
Objective To observe the effects of red clover isoflavones on liver function and inflammatory response in nonalcoholic fatty liver disease rats. Methods Male Sprague-Dawley rats were randomly divided into control group (fed with normal diet), model group (intramuscular fat emulsion) and 18% sucrose solution for 4 weeks. The successful rats were randomly divided into model group (75,150,300 mg / kg red clover isoflavone) were given to the low, middle and high dose red clover groups for 12 weeks. The levels of AST, ALT, Interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-alpha), C-reactive protein (CRP) activity or content. Results Compared with the model group, the hepatocytes in the red clover group had less hepatic injury and improved hepatic function. The contents of serum ALT [(65.63 ± 13.00) U / L] and AST [(145.88 ± 34.28) U / L] (P <0.05). The levels of TNF-α, IL-6 and CRP in middle dose red clover group were (43.24 ± 4.14) ng / mL and (69.94 ± 6.98) pg / mL respectively mg / L, were significantly lower than the model group (P <0.05). Conclusion Red clover isoflavones have a protective effect on liver function in non-alcoholic fatty liver rats, and can reduce the inflammatory reaction.
