目的 观察脑梗死患者血小板表面活化标志物的表达以及血小板膜糖蛋白 (GP)Ibα基因的变异 ,探讨血栓性疾病血小板体内活化与基因多态性的关系。方法 以流式细胞术 (FCM )测定10 2例脑梗死患者血小板GPIb、P 选择素的表达及纤维蛋白原的结合 ,同时应用聚合酶链反应(PCR)技术结合限制性内切酶分析研究GPIbα基因血小板同种抗原 (HPA) 2和可变数目串联重复序列 (VNTR)多态性与等位基因频率分布特点 ,并与相应正常人结果进行比较。结果 脑梗死患者P 选择素及纤原结合明显高于正常组 (P <0 0 1) ,GPIb表达低于正常组 (P <0 0 5 ) ;而GPIbαHPA 2、VNTR基因多态性与对照组相比无明显差异。结论 脑梗死患者体内存在血小板活化 ,但与GPIbαHPA 2及VNTR基因多态性无关。
Objective To observe the expression of platelet surface activation markers and the mutation of platelet glycoprotein (GP) Ibα gene in patients with cerebral infarction and to explore the relationship between platelet activation and gene polymorphism in thrombotic diseases. Methods The expression of platelet GPIb, P-selectin and the binding of fibrinogen in 102 patients with cerebral infarction were measured by flow cytometry (FCM). Meanwhile, the expression of GPIbα and GPIbα were detected by polymerase chain reaction (PCR) and restriction enzyme analysis The genetic polymorphisms of HPA 2 and VNTR and allele frequency distribution were compared with those of corresponding normal subjects. Results The levels of P-selectin and fibrinogen in patients with cerebral infarction were significantly higher than those in the normal group (P <0.01), while those in the GPIb group were lower than those in the normal group (P <0.05). However, the GPIbαHPA 2, No significant difference compared to. Conclusion There is platelet activation in patients with cerebral infarction, but not with GPIbαHPA 2 and VNTR gene polymorphisms.
