HPLC-MS的联用技术当前还处于发展阶段,关键在于接口的研制,美国在这一领域内的研究目前处于领先地位。本文着重介绍美国在高效液体色谱-质谱联用仪的研制、改进以及应用等方面的现状。其中有菲尼根公司作商品出售的机械传送带接口,已达到2ng的检出灵敏度。东北大学的截流萃取器使机械传送接口适应于有缓冲剂的反相层析,用10ng样品获得供鉴定用的谱图。康奈尔大学的直接导入接口达到500pg的检出极限并成功地应用于30—50个氨基酸的多肽分析。H-P公司在探头上作了修改,使直接导入接口实现了商品化。文章还引用35篇文献。6张图、二张表,并对接口的今后研究方向作了介绍。
The key technology of HPLC-MS in combination is still in the developing stage. The key lies in the development of the interface. The research on this field in the United States is currently in the leading position. This article highlights the current status of the development, improvement, and application of HPLC in the United States. Among them are the Finnegan machinery for the sale of goods conveyor interface, has reached 2ng detection sensitivity. Northeastern University’s plug-flow extractor adapted the mechanical interface to buffer-phase reverse-phase chromatography, and 10 ng of sample was used to obtain a chromatogram for identification. The Cornell direct import interface achieves a limit of detection of 500 pg and has been successfully applied to peptide analysis of 30-50 amino acids. H-P company has made changes in the probe, so that the direct import interface to achieve commercialization.
