目的:研究不同浓度的胰岛素对体外培养条件下的健康人外周血内皮祖细胞(endothelial progenitor cells,EPCs)凋亡的影响。方法:经密度梯度离心法分离出健康成年人外周血中的单个核细胞(mononuclear cells,MNCs),在体外诱导分化后,用Dil-acLDL和FITC-UEA-1双染色鉴定为内皮祖细胞。观察不同浓度的胰岛素对将所获取的内皮祖细胞凋亡的影响。结果:20mU/L胰岛素干预组较对照组的的凋亡率差异无统计学意义(P>0.05);200mU/L及2000mU/L胰岛素干预组较对照组的凋亡率差异有统计学意义(P<0.05),且2000mU/L胰岛素干预组的凋亡率随着干预时间的延长而增加(P<0.01)。结论:高浓度胰岛素可增加体外培养的人外周血内皮祖细胞的凋亡,并且这种影响呈时间依赖性。
Objective: To study the effects of different concentrations of insulin on the apoptosis of peripheral blood endothelial progenitor cells (EPCs) in healthy volunteers. Methods: Mononuclear cells (MNCs) were isolated from peripheral blood of healthy adults by density gradient centrifugation. After induced differentiation in vitro, they were identified as endothelial progenitor cells by double staining with Dil-acLDL and FITC-UEA-1. To observe the effect of different concentrations of insulin on the apoptosis of the obtained endothelial progenitor cells. Results: The apoptosis rate of 20mU / L insulin intervention group was not significantly different from that of the control group (P> 0.05). The apoptosis rates of 200mU / L and 2000mU / L insulin intervention group were significantly different from those of the control group P <0.05), and the apoptosis rate of 2000mU / L insulin intervention group increased with the prolongation of intervention time (P <0.01). Conclusion: High concentration of insulin can increase the apoptosis of human peripheral blood endothelial progenitor cells cultured in vitro, and the effect is time-dependent.
