构建过表达整合蛋白α5β1的SMMC7721细胞株。方法应用分子克隆技术和基因转染建立过表达整合蛋白α5β1的SMMC7721细胞株,应用RT-PCR及流式细胞术鉴定SMMC7721细胞表面整合蛋白α5β1的表达。结果获得过表达整合蛋白α5/β1的SMMC7721细胞株α5.3-7721,β1.6-7721,α5β1.6-7721细胞。结论整合蛋白α5β1基因转染后,SMMC7721细胞整合蛋白α5β1的RNA及蛋白质表达明显提高。过表达整合蛋白α5β1的SMMC7721细胞株的建立有助于研究整合蛋白α5β1在肝癌中的作用
Construction of SMMC7721 cell line overexpressing integrin α5β1. Methods The SMMC7721 cell line overexpressing integrin α5β1 was established by molecular cloning and gene transfection. The expression of integrin α5β1 on SMMC7721 cells was identified by RT-PCR and flow cytometry. Results SMMC7721 cells overexpressing the α5 / β1 integrin α5.3-7721, β1.6-7721 and α5β1.6-7721 cells were obtained. Conclusion The expression of integrin α5β1 mRNA and protein in SMMC7721 cells after transfection of integrin α5β1 gene was significantly increased. The establishment of SMMC7721 cell line overexpressing the integrin α5β1 contributes to the study of the role of integrin α5β1 in hepatocellular carcinoma
