目的探讨20-羟基蜕皮甾酮(20E)对糖尿病肝病的保护作用及相关机制。方法大鼠(体重80~100 g)随机分4组,用60 mg/kg腹腔注射链脲佐菌素(STZ)造模,之后正常对照组与模型组灌服蒸馏水,阳性对照组和20E治疗组分别灌服5 mg/kg盐酸二甲双胍和10 mg/kg 20E溶液。药物干预6周后,测定血糖、糖原、胰岛素、丙转氨酶(ALT)、谷草转氨酶(AST)和血糖(Glu)等含量,分析促炎性细胞因子肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、干扰素-γ(interferon-γ,INF-γ)和环氧合酶2(COX-2)的表达。结果与模型组相比,20E处理后肝脏损伤程度减轻,Glu、ALT、AST和ALP含量分别降低了62.4%(P﹤0.01)、52.5%(P﹤0.05)、43.7%(P﹤0.05)和49.1%(P﹤0.05),糖原和胰岛素含量增加了152.3%(P﹤0.05)和96.4%(P﹤0.05),TNF-α、IL-6、IN-γ和COX-2mRNA水平分别降低了68.2%(P﹤0.05)、56.1%(P﹤0.05)、6.6%和30.2%(P﹤0.05)。结论 20E对糖尿病肝病具有保护作用,其机制可能与降低Glu、ALT、AST和ALP含量,增加糖原和胰岛素浓度和下调TNF-α、IL-6、IN-γ和COX-2表达有关。
Objective To investigate the protective effect of 20-hydroxy ecdysterone (20E) on diabetic liver disease and its related mechanism. Methods Rats (body weight 80-100 g) were randomly divided into 4 groups, which were injected intraperitoneally with streptozotocin (STZ) at a dose of 60 mg / kg. The normal control group and model group were given distilled water, positive control group and 20E treatment Group were fed with 5 mg / kg metformin hydrochloride and 10 mg / kg 20E solution. Six weeks after the drug intervention, contents of blood glucose, glycogen, insulin, ALT, AST and Glu were measured to detect the levels of tumor necrosis factor-α (TNF-α), proinflammatory cytokines Interleukin-6 (IL-6), Interferon-γ (INF-γ) and Cyclooxygenase 2 (COX-2) Results Compared with the model group, the liver damage was alleviated by 20E, the levels of Glu, ALT, AST and ALP were decreased by 62.4% (P <0.01), 52.5% (P <0.05), 43.7% The levels of glycogen and insulin increased by 152.3% (P <0.05) and 96.4% (P <0.05), while the levels of TNF-α, IL-6, IN-γ and COX- 68.2% (P <0.05), 56.1% (P <0.05), 6.6% and 30.2% respectively (P <0.05). CONCLUSION: 20E has a protective effect on diabetic liver disease. The mechanism may be related to decreasing Glu, ALT, AST and ALP, increasing glycogen and insulin concentration, and decreasing TNF-α, IL-6, IN-γ and COX-2 expression.
