应用二种中性盐溴化红和氯化钙作为溶解蚕丝丝素的试剂,制成了二种固定化葡萄糖氧化酶丝素膜。经酶比色法和红外分光光度法分析,结果表明这二种丝素膜都是良好的固定化酶的生物材料。葡萄糖氧化酶经这些丝素膜固定以后对热和pH稳定性得到明显改良。这些酶膜性能稳定,具有高的活性得率,能长期存放。以这些固定化葡萄糖氧化酶丝素膜和氧电极为基础,研制的流动注射分析式电流型葡萄糖生物传感器性能较稳定,具有较宽的葡萄糖线性响应范围(0.5~15.0mmol/L),相关系数为0.999,可以反复测定葡萄糖一千次以上。
Two kinds of neutral salt bromide red and calcium chloride were used as reagents to dissolve silk fibroin, two kinds of immobilized glucose oxidase silk fibroin films were made. The enzyme colorimetric and infrared spectrophotometric analysis showed that the two kinds of silk fibroin membrane are good biomaterials for immobilized enzymes. Glucose oxidase has been markedly improved in heat and pH stability after fixation with these silk fibroin films. These enzyme membranes are stable, have high activity yields and are long-term storage. Based on these immobilized glucose oxidase silk membrane and oxygen electrode, flow-injection current glucose biosensor developed more stable, with a wide range of glucose linear response (0.5 ~ 15.0mmol / L), the correlation coefficient 0.999, you can repeatedly measure more than one thousand times glucose.
