目的建立了固相萃取-高效液相色谱同时测定蛋白粉中6种合成色素的检测方法。方法样品经Pro Elut PLS固相萃取柱净化,浓缩,通过Diamonsil 5μm C18反相色谱柱分离,以乙腈-乙酸铵缓冲溶液为流动相进行梯度洗脱,采用二极管阵列检测器检测。结果各色素组分在1~50 mg/L范围内线性良好,回归系数均大于0.999 6,检出限为0.03~0.10 mg/L;蛋白粉样品中6种色素的回收率为83.8%~101.3%,RSD为0.84%~3.25%。结论该方法简便、快速、分离效果好、灵敏度高、重现性好,适用于蛋白粉样品中合成色素的测定。
OBJECTIVE To establish a method for simultaneous determination of six synthetic pigments in protein powder by solid phase extraction and high performance liquid chromatography. Methods The samples were purified by Pro Elut PLS SPE cartridge, concentrated and separated by Diamonsil 5μm C18 reversed-phase column. The mobile phase was eluted with acetonitrile-ammonium acetate buffer solution and detected by diode array detector. Results The results showed that the linearity of each pigment component in the range of 1 ~ 50 mg / L was good and the regression coefficients were all greater than 0.999 6 with the detection limits of 0.03 ~ 0.10 mg / L. The recoveries of six pigments in protein powder samples were 83.8% ~ 101.3 %, RSD 0.84% ~ 3.25%. Conclusion The method is simple, rapid, good separation, high sensitivity and good reproducibility. It is suitable for the determination of synthetic pigment in protein powder samples.
