目的:比较小鼠免疫共刺激信号B7-1和B7-2转化小鼠淋巴瘤细胞株EL4后,转化细胞介导的抗肿瘤免疫作用。方法:构建逆转录病毒表达载体pLXSN-mB7-1和pLXSN-mB7-2,转染野生型EL4细胞,获得高表达B7-1和B7-2分子的EL4/mB7-1和EL4/mB7-2细胞,并制备成瘤苗;分别应用LDH释放法、MTT法以及ELISA法检测比较两种瘤苗激活的T细胞的杀伤活性、细胞增殖和IL-2的分泌情况。结果:①EL4/mB7-1和EL4/mB7-2瘤苗刺激小鼠脾脏T淋巴细胞分泌IL-2的量明显超过野生型EL-4细胞(P<0.01);②24 h,EL4/mB7-1瘤苗刺激小鼠脾脏T淋巴细胞分泌的IL-2的量超过EL4/mB7-2瘤苗(P<0.05);48 h,EL4/mB7-2瘤苗刺激小鼠脾脏T淋巴细胞分泌IL-2的量超过EL4/mB7-1瘤苗(P<0.05);③EL4/mB7-1瘤苗和EL4/mB7-2瘤苗刺激小鼠T淋巴细胞杀伤肿瘤细胞的活性明显增强。EL4/mB7-2瘤苗刺激小鼠T淋巴细胞杀伤肿瘤细胞的活性与EL4/mB7-1瘤苗相当(P>0.05)。结论:mB7-1和mB7-2分子可活化T细胞分泌IL-2,两者刺激T细胞产生IL-2的高峰时间不同。mB7-2刺激T细胞产生CTL的杀伤活性与mB7-1无明显差别。
OBJECTIVE: To compare the effects of mouse immunostimulatory signals B7-1 and B7-2 on the transformation of murine lymphoma cell line EL4 into transformed cell-mediated antitumor immunity. Methods: The retroviral vectors pLXSN-mB7-1 and pLXSN-mB7-2 were constructed and transfected into wild-type EL4 cells to obtain EL4 / mB7-1 and EL4 / mB7-2 highly expressing B7-1 and B7-2 Cells and prepared into tumor-bearing vaccine. The killing activity, cell proliferation and IL-2 secretion of T cells activated by two kinds of tumor vaccine were compared by LDH release assay, MTT assay and ELISA assay respectively. Results: ① The amount of IL-2 secreted by T lymphocytes in mice spleen was significantly higher than that in EL-4 cells (P <0.01) by EL4 / mB7-1 and EL4 / mB7-2 vaccine; The amount of IL-2 secreted by spleen T lymphocytes of mice was more than that of EL4 / mB7-2 tumor vaccine (P <0.05) after 48 hours. The splenic T lymphocytes stimulated by EL4 / mB7-2 vaccine stimulated IL- 2 over EL4 / mB7-1 tumor vaccine (P <0.05). (3) EL4 / mB7-1 vaccine and EL4 / mB7-2 vaccine stimulated the T lymphocytes of mice to kill tumor cells significantly. The activity of EL4 / mB7-2 vaccine in killing T lymphocytes in mice was similar to EL4 / mB7-1 vaccine (P> 0.05). Conclusion: The mB7-1 and mB7-2 molecules can activate T cells to secrete IL-2, and both of them stimulate the T cells to produce different levels of IL-2. The cytotoxic activity of mB7-2 stimulated T cells to CTL showed no significant difference from that of mB7-1.
